dcterms.abstractIntroduction: Extensive clinical studies on meridian high-voltage electrical stimulation (Dao-ping) (經絡導平治療) demonstrated that Dao-ping could restore the motor functions from stroke-induced hemiplegia, improve the facial actions from facial paralysis, and reduce the pain level of sciatica resulted from stroke, and recover from frozen shoulder. Significance: It remains unclear if Dao-ping could be used as a measure for stroke prevention. Methods: A total of 120 male Sprague-Dawley rats (300g-350g) were used. Pre-ischemia Dao-ping treatment was given to normal rats for 5 times or 18 times daily at bilateral Zusanli (ST36), or Shousanli (LI10), or pair of them, in which both acupoints were commonly, clinically used in patients with stroke treated with Dao-ping. After multiple Dao-ping treatment, transient focal cerebral ischemia was induced surgically. The right middle cerebral artery was exposed and occluded by surgical clip for 1 hour. then released for reperfusion. The survival rate of rats and the lipid peroxidation assay of brain tissue were measured. Results: The overall survival rate of cerebral ischemic rats after Dao-ping treatment was high before harvesting reflected its safety on animals. In testing the acupouints, The 5-times and 18-times pre-ischemia Dao-ping-ST36 were compared on ischemia rats (n=5), the amount of Malondialdehyde (MDA) only and Malondialdehyde (MDA) + 4-hydroxy-2(E)-nonenal (4-HNE) the amount of lipid peroxidation was significantly reduced by 24.07% to 12.65 ± 2.84 uM Malondialdehyde (MDA) only per mg protein (p<0.05), and significantly reduced by 19.82% to give 14.89 + 1.84 uM Malondialdehyde (MDA) + 4-hydroxy-2(E)-nonenal (4-HNE) per mg protein (p<0.05). When 18-times pre-ischemia Dao-ping-LI10 was applied on ischemia rats (n=5), the amount of lipid peroxidation was also significantly reduced by 17.11% to 13.81 ± 2.74 uM MDA per mg protein (p<0.05), and significantly increased by 10.02% to give 16.71 ± 3.95 uM MDA + 4-HNE per mg protein (p<0.05). When 18-times pre-ischemia Dao-ping-LI10 and ST36 was applied on ischemia rats (n=5), the amount of lipid peroxidation was significantly reduced by 13.27% to 14.45 ± 2.03 uM MDA per mg protein (p<0.05), and significantly reduced by 15.40% to give 15.71 ± 1.15 uM MDA + 4-HNE per mg protein (p<0.05). Conclusions: The survival rate of cerebral ischemia rats with Dao-ping stimulation was high before harvesting. The results of the study provided direct evidence of the efficacy of Dao-ping. The effect is significant on ST36, but not on LI10.en_US
