Abstract: | A subset of Adenoid Cystic Carcinoma (ACC) is characterized by MYB fusions including the novel t(6;9)(q22-23; p23-24) translocation. There has been over expression in MYB RNA in ACCs regardless to MYB fusions. Retrospective study of formalin-fixed paraffin embedded (FFPE) samples of 21 salivary gland ACCs and 3 lacrimal gland ACCs together with 6 non-ACC salivary gland tumors were studied. As a result, MYB-NFIB fusion transcripts in salivary gland adenoid cystic carcinoma and lacrimal gland carcinoma were 9% and 50% respectively with an overall frequency of 15%. No MYB-NFIB fusion transcripts were identified in all non-ACC of salivary gland, suggestive of a high specificity of MYB-NFIB fusion in ACC of salivary gland. Sixty-seven percent of salivary gland ACC were MYB fusion positive. In case of lacrimal gland ACC, all cases were fusion positive. The overall frequency of MYB fusions was therefore 71%. MYB expression was found in 80% and 67% of fusion-positive salivary gland and fusion-positive lacrimal gland ACCs and was about 67% of fusion-negative salivary gland ACCs despite of low expression in fusion-negative ACCs. MYB over expression didn't appear in other salivary gland tumors. Overall percentage of MYB expression in ACC samples was 75% whereas the percentages of MYB expression in fusion positive and negative ACC samples were 78% and 67% respectively. Hence, MYB can be counted as a major oncogene in ACC pathogenesis. |
