Abstract: | Introduction: Alzheimer's disease (AD) is the most common form of dementia, which account for 60-80% of dementia case. One-tenth of the elderly population in Hong Kong was suffered from AD and that adds burden to the healthcare system in Hong Kong. The disease is characterized by behavioral changes and memory loss, along with physiological changes such as atrophies in brain regions. The mechanism of AD pathogenesis had been extensively studied but no definitive factor was confirmed. Oligodendrocytes belongs to glial cells in the CNS. The myelinated layer of oligodendrocytes is responsible for transmission of signal and structural integrity of the whole neural network. Dysregulation of oligodendrocytes was found in various neurodegenerative diseases, including AD. On the other side, the apolipoprotein E4 (APOE4) allele is considered as the greatest risk factor in the disease development of late on-set AD. The relationship between possessing APOE4 allele and dysregulation of oligodendrocytes are remain unclear. Material and Method: APOE4 and APOE3 (human normal control) transgenic mice were used in this study. Brain tissues sections were stained with H&E and CV for histological examination. IHC staining of sections with oligodendrocytes and OPCs specific markers were performed. The images of IHC slides were analyzed by bioimaging software QuPath to obtain positive cell number and cellular density data. Reverse transcription quantitative PCR was performed to investigate the mRNA expression level of myelin related genes and lipid metabolism related gene. Result: Structural differences were detected in both H&E and CV-stained brain tissue. IHC results reveal that both percentage and density of oligodendrocytes and OPCs were reduced in hippocampus of APOE4 mice, while the results of the other regions were either unchanged or reduced. The qPCR reveal that myelin related genes were upregulated in APOE4 mice while the results of lipid metabolic genes were undetermined. Conclusion: Findings suggested that oligodendrocytes and OPCs density is reduced in APOE4 mice, while myelin related genes were upregulated. Further studies are suggested to understand the mechanism behind this contradictory result. |
