dcterms.abstract | In this study, Alcaligenes latus DSM 1122, 1123, 1124 from Tsinghua University, Beijing and bacteria from activated sludge were utilised to produce polyhydroxyalkanoates (PHA). In the first part of the study, a mixed culture in activated sludge was used to produce PHA by Sequencing Batch Reactor. The nutrient for bacteria growth stage was hydrolysed soya waste. Glucose was supplied as a carbon source for the PHA accumulation stage. PHA polymer yield was 0.20 g which could be obtained from 1 kg soya waste and 176 g glucose. The PHB: PHV copolymer ratio of the polymer was 79:21 (w/w). The second part of study was conducted by experimenting different conditions of treatment on the malt refuse as the nutrient, and usage of A. latus DSM 1122, 1123, 1124 and bacteria isolated from sludge to produce PHA. Shaking flask method was employed. The result showed that 9 mg and 25 mg polymer could be produced from 100 g malt waste by DSM 1122 and 1124 respectively. And both of the bacteria produced PHB homopolymer from malt extract. Identification and the antibiotic sensitivity test of the bacteria isolated from activated sludge were carried out and this bioplastic productive bacteria was known as Klebsiella pneumoniae. | en_US |
