dcterms.abstract | The major aim of this project is to study the applicability of scientific methodologies, i.e., elemental, infrared and proteomic analysis, on Ling Zhi (Ganoderma Lucidum) for analysis and identification purposes. A method for determining major (Al, Ba, Ca, Fe, Na, Mg, K, Sn, Sb and P) and trace (V, As, Se, Cd, Hg, Cu, Cr, Ni, U and Pb) elements in Ling Zhi by inductively coupled plasma-atomic emission spectrometer (ICP-AES) and inductively coupled plasma-mass spectrometer (ICP-MS) after microwave digestion was setup. A Dynamic Reaction Cell (DRC) was used to remove polyatomic interference in the analysis of As and Se at m/z75 and 78, respectively. Method development and validation was accomplished through the use of a standard reference material, SRM 1547, peach leave. Results for the reference samples agreed satisfactorily with certified values. The method was then applied to the analysis of Ling Zhi. The lowest limit of detection for major and trace elements are in the range of hundreds of ppt to sub-ppt level. The results show that the concentration of toxic elements in the samples tested is low and they are safe for consumption. Also, it provides a generous amount of Ca, Mg, P and Zn for our daily need. Infrared and near infrared spectra were scanned and the spectra were compared. The comparisons were base on the absorption at each wavelength. The correlation coefficient of the spectra was calculated with the application of several mathematical filters to correct noise, water and carbon dioxide interference. In FTIR analysis, the correlation coefficients of different part of Ling Zhi fell within 1 to 0.9 where the correlation coefficient of Ling Zhi and mushroom was about 0.6. Although spectra of different parts of Ling Zhi showed no significant difference, spectra of Ling Zhi and mushroom can be differentiated. The use of FTIR spectroscopy to verify the existence of Ling Zhi and other plants is therefore possible. Two dimension polyacrylamide gel electrophoresis (2D-PAGE) was used to separate proteins present in different parts of Ling Zhi. Lysis buffer and TCA/acetone sample extraction methods were used. The results showed that TCA/acetone method extracted more proteins than Lysis buffer extraction. The 2D-PAGE chromatograms of different parts of Ling Zhi showed different patterns. Therefore, 2D-PAGE may be an appropriate method for the protein separation and the differentiation of different parts of Ling Zhi. Also, there were some similarity regions in the chromatograms. It suggested that different parts of Ling Zhi have certain degree of similarity in proteins expression. | en_US |
