Abstract: | In order to explore the feasibility of vegetative propagation of Bauhinia blakeana using plant tissue culture technique, callus initiation was conducted using node, internode and leaf blade samples and compared with another two common Bauhinia species in Hong Kong, B. pururea and B. variegata. Callus samples were successfully induced in B. blakeana and B. variegata. They grew best, in terms of cell proliferation, in MS medium having the concentrations of 1.0 mg/L BA and 2.0 mg/L NAA. No callus was successfully induced in B. pururea, which might due to high contamination rates by endogenous fungi. Root and/or shoot deferentiation was attempted in callus samples of B. blakeana using different hormone combinations of auxins (IAA, IBA, 2,4-D, NAA), cytokinins (BA, kinetin) and Giberellic acid (GA3). However, no successful case were obtained. Pollen germination study was conducted to explore the possible causes of the infertility in B. blakeana. The germinated rate of B. blakeana pollen is generally low, less than 5%. The inhibitory effects of heavy metal on pollen germination was tested. 20 mg/L of Ni2+ and Cu2+ showed complete inhibition on pollen tube germination. 5 mg/L of Ni2+ or Cu2+ showed slightly inhibitory effects on pollen tube germination but still pollen tubes were induced respectively within the 4 and 6-hr of investigation. The results suggested that pollens of B. blakeana showed normal physiological response towards suitable medium and chemical inhibition. However, the ease and successful rate of fertilization of egg cell which is the first step for the success in fruit bearing, is yet to be explored. |
