dcterms.abstract | This thesis demonstrates the use of nanoporous alumina membranes for diffusion of some protein based biomolecules, including bovine serum albumin (BSA), insulin and one kind of anti-cancer drug, cisplatin (CDDP). With the construction of a polydimethylsiloxane (PDMS) chamber, the nanoporous membrane having 20 nm and 100 nm diameter is used as a diffusion controlling system. The diffusion properties are being investigated and compared with the two membrane pore size. The three biomolecules with different concentration is placed in to the chamber with a "filter" of the membrane, and allowed sufficient time for diffusion from one side to another side. The diffusion properties can be monitored by UV-visible spectroscopy since protein has maxima of absorption peak in 280 nm wavelength and CDDP has a range between 280 nm to 300 nm. The results would show the relationship of diffusion rate versus to nanopore size of the membrane and the concentration of certain biomolecules. To further investigate the diffusive effects of the biomolecules, some impedance testing have been done with the cancer cell line KYSE30 (from human esophageal squamous epithelial cancer cell). In the whole experiments, it herein serves as an exploratory test for the application of nanoporous alumina in novel drug delivery system. | en_US |
