dcterms.abstract | Cancer is a major cause of death worldwide. Despite the availability of various cancer therapies and the resulting cancer remission, relapse still occurs. A plausible explanation to this is the cancer stem cell (CSC) hypothesis. The cancer stem cell hypothesis suggests that a minority of cells are tumorigenic. They also possess multidrug resistance (MDR). It is therefore important to have drugs that can selectively target CSCs. This MDR feature is believed to be attributed by the ABC transporters. In spite of that, they are very useful in identifying CSCs by Hoechst staining in vitro. In view of this, we utilized Hoechst 33342 staining technique to investigate the effect of a clinically used anti-tumor TCM Huachansu, on the side population (SP) of 2 cancer cell lines (KYSE150 & A549). First of all, a platform to study SP was optimized by studying the growth rate of cells, identifying the Hoechst 33342 concentration and evaluating the toxicity effect of HCS on the cells. Subsequently, the cells were treated with different concentrations of HCS and the SP percentages were measured. It was found that both cell lines showed a dosage effect toward HCS. HCS treatment is statistically significant in SP% reduction in KYSE150 (p<0.05). The post hoc comparison by LSD suggested that HCS 1/30 & 1/10 produced significant differences in SP% compared to the untreated group (i.e.PBS).
