dcterms.abstractBackground: Chinese herbal medicine with promoting blood circulation and removing blood stasis properties, are commonly used for prevention and treatment of cardiovascular and cerebrovascular diseases. These herbs may possess the anticoagulant effect by inhibiting the formation of blood clot or thrombi. Aim of the study: The antithrombotic effect of Danshen, Honghua, Taoren and Yi Mu Cao are commonly applied as Traditional Chinese herbal medicine to treat and prevent thrombi. Their effects on blood coagulation were evaluated in normal and warfarinized citrated plasma samples with different concentrations of herbal solutions in order to determine the dose.dependent effect. Materials and methods: The in vitro effect of Danshen, Honghua, Taoren and Yi Mu Cao were determined in normal and warfarinized citrated plasma samples by measuring the prothrombin time (PT) and activated partial thromboplastin time (APTT). The coagulation tests were done by using Sysmex Automated Coagulation Analyzer CA-500 with the Thromborel S reagent for measuring the PT while Actin FSL and calcium chloride for detecting the APTT. Each citrated plasma samples were separated into 4 single tubes and mixed with different concentrations of herbal working solution including control group, therapeutic dose, high-dose and double high-dose to evaluate the effect of herbs on blood coagulation. The final concentration of therapeutic dose, high-dose and double high-dose of the selected herbs in plasma concentration were tested in the study with 6 mg/mL, 15 mg/mL, 30 mg/mL for Danshen; 3 mg/mL, 6 mg/mL, 12 mg/mL for Honghua; 2 mg/mL, 4 mg/mL, 8 mg/mL for Taoren; 6 mg/mL, 30 mg/mL, 60 mg/mL for Yi Mu Cao.en_US
dcterms.abstractResults: Danshen, Honghua and Yi Mu Cao significantly delayed the PT and APTT among normal and warfarinized citrated plasma samples in therapeutic dose, high dose and double high-dose of herbs as compared to control group. However, there was no significantly difference in PT in the normal plasma groups among different dosages of Taoren. Taoren significantly shortens the PT in warfarinized plasma among different dosages. On the other hand, Taoren only prolonged the APTT in normal and warfarinized plasma with high-dose and double high-dose of Taoren. Conclusion: The usage of Danshen, Honghua, and Yi Mu Cao was found to be able to inhibit the coagulation process through prolonged PT and APTT. The results were showed the inhibitory effect on the blood coagulation for properties of promoting blood circulation and removing blood stasis in concept of TCM. However, patients on warfarin therapy should avoid taken with Danshen, Honghua, and Yi Mu Cao together as it may increase the risk of bleeding tendency. Further investigation for hemorheology of Chinese herbal medicine is needed to provide exploration and evidence-based references for herb safety and toxicity concerned so that Chinese herbs can be an alternative antithrombotic agent to replace the use of warfarin.en_US
