dcterms.abstractDNA damage is a crucial factor in ageing, which in turn contributes the development of age-related disease. Evidence shows that diets high in fruits and vegetables can diminish oxidative damage to DNA and protect against cancers and chronic diseases. Concord grape juice from the grapes of Vitis labrusca have been investigated for its antioxidative power and its potential health benefits. Protective effect of single dose of concord grape juice on human lymphocytic DNA was examined. In an unblinded, controlled, crossover single dose study, ten healthy volunteers took 300 mL commercial 100% concord grape juice or vitamin C and glucose in drinking water (the control drink). Subjects took all the treatments in random order with two weeks’ washout between each treatment. Two mL venous blood was taken before and 2 hour after drinking in each occasion. Lymphocytes were isolated from the venous blood and DNA damage was measured by the standard comet assay. Levels of DNA damage in concord grape juice supplementation were compared with control drink. The first part of this study was to investigate the in vivo effect of concord grape juice consumption on basal DNA protection. DNA protective effect was further tested after 2 hours of concord grape juice intake followed by oxidative stress mediated by hydrogen peroxide (H₂O₂). Our data showed that significant decrease of 24% baseline DNA damage after grape juice consumption under unstressed condition (p < 0.05). Significant DNA protective effect was seen in H₂O₂ induced oxidative stressed cells after two hours acute ingestion with grape juice (p < 0.05). Furthermore, the comet assay results showed 49% decrease in DNA damage with concord grape juice acute ingestion. Such effect was not seen in consumption of control drink. Intake of concord grape juice offered a speedy DNA protective effect on human DNA. We have demonstrated antioxidant effect of concord grape juice in vivo, not attributed entirely to the vitamin C content of the juice. This protective effect may offer lower oxidative damage to DNA and potentially protect against certain age-related diseases.en_US
