dcterms.abstractLung cancer is the leading cause of cancer death in the world. Despite there are various therapies, the relapse of lung cancer is still occurring. This means that traditional therapies are not enough or become ineffective. A new subset of cells in cancer which is called cancer stem cells (CSCs) had been demonstrated to contain the abilities of self-renewal, differentiation and multi-drugs resistance. Therefore, CSCs becomes as the new target for cancer therapy. Aldehyde dehydrogenases (ALDH) had been defined as the marker for CSCs. Measurement of ALDH in cancer cells can be used for testing the effects of the drug, which is specific target on CSCs. Rhizoma Paridis saponins (RPS) which is the Traditional Chinese Medicine (TCM) had been widely used for the treatment of lung cancer. However, the anti - cancer effect to CSCs in lung cancer had not been studied. In view of this, the effect of RPS to CSCs by detecting ALDH in A549 cell line that is the non-small cell lung cancer was investigated by using flow cytometer. In order to detect the CSCs in A549 cell line, the platform for measurement of ALDH had been established and optimized. The reduction effects to ALDH% cells from RPS were from 53% to 87% (p-value<0.05) when the concentrations of RPS were increased (from 125ug/ml to 1000ug/ml). Furthermore, RPS also contained the cytostatic effect to lung cancer by reducing the numbers of cancer cell but retained high viability. In conclusion, RPS was demonstrated to contain the effect for decreasing the CSCs in non-small cell lung cancer.en_US
