dcterms.abstract | Taurine had been well known to be involved in many physiological processes in order to prevent diabetes, hypertension, inflammatory diseases and also for neuroprotection. The antioxidant role of taurine was found to have potential anti-cancer effects based on inhibition of tumor proliferation and angiogenesis with enhancement of tumor cell apoptosis. Certain studies found that significant reduction of plasma taurine levels were shown on cancer patients when compared to normal. It was hypothesized that taurine treatments could help prevent and cure of cancers. In this study, wooden tips ESI-MS assay was developed for plasma taurine quantification, in order to investigate the anti-cancer effects of taurine using mouse cancer models. Total of 29 mice plasma samples were obtained from SZRI, in which 14 and 15 samples were come from cancer prevention and treatment models respectively. They were collected from specific pathogen free (SPF) grade male, aged 6-8 weeks C57BL/6J mice with intact immune system and inoculated with B16 melanoma cells. Each model was further subdivided into 3 groups, including Vehicle (control), T (treatment of taurine) and G+T (treatment with taurine and ganoderma). Taurine levels were compared between groups after different treatments for both models. Then the correlation between taurine levels and provided tumor sizes was studied in order to evaluate the effective of treatments. The results showed that there were possible ion suppression and matrix interference when using wooden tips for electrospray ionization, narrowing the linear ranges and reducing the accuracy as well as precision. In addition, the overall findings did not indicate elevation of taurine levels after different treatments, based on dysfunction of taurine reabsorption in C57/BL6J mice. There were also no significant correlations of taurine levels and tumor weights after treatments for both models. It would be due to limitation of linear regression and also small sample sizes. It is suggested to use hydrophobic or semi-polar polyester spraying tips instead of wooden tips, and perform survival analysis by comparing the possibility of survival between different treatments. | en_US |
