Abstract: | Introduction: With better understanding of the molecular mechanism of vitamin D, evidence showing its non- skeletal roles and its relationship with adverse health outcomes has been rapidly expanding. The linkage between oxidative stress and vitamin D status has been of particular interest because of the suggested effect of vitamin D in lowering oxidative stress, which may consequently lower the risk of disease outcomes which are closely linked to oxidative stress.Objective: In this study, the relationship between vitamin D status and oxidative stress in a group of apparently healthy young adults was investigated. Method: Apparently healthy volunteers (28 female and 14 male, aged 18- 24) were recruited with their informed consent, and the levels of 25- hydroxyvitamin D3 in plasma and those of the oxidative stress biomarkers 8- oxo- 7, 8- dihydro- 2'- deoxyguanosine (8-oxodG), 8- dihydroguanosine (8- oxoGuo) in urine and allantoin in plasma were measured using liquid chromatography- tandem mass spectrometry. Urinary8- oxoGuo and 8- oxodG concentrations were expressed in relation to urinary creatinine concentration. Strength of correlation was investigated using Spearman rank correlation at significance level p = 0.05 Results: Mean (SD) results for plasma 25(OH) D were 38.34(12.9) nmol/l; for plasma allantoin, 4.31(1.65) mmol/l, and for urinary 8- oxoGuo and 8oxodG were, respectively, 2.41(1.50) and 1.93(1.15) nmol/ mmol creatinine. No significant correlation was observed between plasma 25(OH) D and any of the biomarkers of oxidative stress measured: (25(OH) D and allantoin: r= 0.06, p= 0.97; 25(OH) D and 8- oxodG: r= 0.008, p= 0.96; 25(OH) D and 8- oxoGuo: r= 0.069, p= 0.70) Conclusion: This study does not show any correlational relationship between the level of 25 (OH) D and allantoin, 8- oxodG and 8- oxoGuo respectively in apparently healthy young population. Further investigation with larger number of participants and careful selection of subject according to their vitamin D level is suggested. |
