|Abstract:||Traditional method to study muscle force and the relationship between muscle force and other factors such as the intracellular calcium ion concentration relies on electric signals, while at present it cannot test the muscle forces without a pre-determined model. Facing these limitations, new method for muscle measurement has been one research hotspot. Eight frog samples were used for tested. The frogs have experienced the same cultivation and fed. Also when sacrificing and sample processing, all steps are following the standard guidance and all samples experienced the same processing. The samples have been calmed, then cultivated, then warmed, then dyed. The Fura-2, one widely-used fluorescence probe, and a set of equipment of testing Fura-2 were used to conduct the experiment and the tension is tested on the standard frog plate controlled by the computer. Also sensitive micro sensor has been used to measured calcium concentration at rest state. As expected, the main parameter of Fura-2, the ratio of fluorescence strength excited under 380nm and 344nm wavelength can change with the tension produced by stimulations. The synchronism has been shown that the Fura-2 can be used for intact muscle tissue tension measurement. Also the intracellular concentration has been estimated by Fura-2, at rest is 115.1±7.5 nmol/L, and 110.3±3.4 nmol/L measured by calcium sensitive micro sensor. Meanwhile the calcium concentration at 50% tension ([Ca2+]50 for short) for 8 samples is 692.8±11.7 nmol/L, and the tension at 50% is 1.9±0.2 mN. The correlation coefficient between [Ca2+]50 and 50% tension is -0.041. A nonparametric test shows that here is a significant result between [Ca2+]50 and 50% tension with a p=0.016.|
